I have been a member of the Mandrup research group since the spring of 2022, where I joined as a bachelor student. I investigated the role of PPARd during adipogenesis using 3T3-L1 preadipocytes as a model system. A year later in 2023 I did an individual study activity (ISA) focusing on investigating the transcriptional regulation of leptin. Here I identified two putative enhancers around the leptin gene in both human and mouse. This led to my master where I continued to investigate the transcriptional regulation of leptin. I used knockout mouse models to investigate the importance of the two putative leptin enhancers. Additionally, I used a human cell strain to uncover the transcriptional regulation of the human leptin gene and identify dynamically regulated regions in the leptin locus by using pharmacological activators and repressors. Currently I am employed as a research assistant in the Mandrup Group to continue the investigation with focus on the two putative enhancers in mouse models.
